Verdict. Cholesterol that HDL delivers through SR-B1 leaves the plasma membrane, lands in the ER, and that sterol pool is what assembles mitochondria-associated membranes. The contacts then esterify surplus cholesterol and turn down biosynthesis, so extracellular lipoproteins set the ER-mitochondria interface.
What the authors report
Mitochondria-associated ER membranes (MAM domains, or MAMs) are transient lipid-raft domains of the ER that coordinate metabolic pathways and ER-mitochondria communication. How they form has been poorly understood. The authors show that MAM assembly is dynamically regulated by plasma-membrane cholesterol. Cholesterol delivered through high-density lipoproteins via scavenger receptor class B type 1 is rapidly redistributed from the plasma membrane to the ER. Increased cholesterol availability there promotes MAM assembly. In turn, MAM formation promotes communication between the ER and mitochondria, esterification of excess cholesterol, and suppression of cholesterol biosynthesis. Disrupting MAMs uncouples those responses and alters trafficking and metabolic adaptation to extracellular cholesterol. They call MAM a dynamic metabolic regulator that translates lipoprotein supply into intracellular adaptation.
What was measured
HDL/SR-B1 cholesterol delivery, plasma-membrane to ER redistribution, MAM assembly, ER-mitochondria communication, cholesterol esterification, and biosynthetic suppression. The abstract does not name cell type, MAM tethers, or the esterifying enzyme.
Caveats
Abstract only. Preprint. Esterification is reported without naming ACAT or any other enzyme. The MAM disruption method is unspecified. No OXPHOS, membrane-potential, or metal-site measurement is claimed.
What to do with it
Treat MAM number and function as cholesterol-gated by HDL/SR-B1, not as a static architecture. The molecular test is whether blocking SR-B1 uptake, or lowering ER cholesterol, prevents MAM assembly and the esterification/biosynthesis couple when extracellular HDL rises.
