IOMM-Lee is an NF2-wildtype, TERT-mutant Grade 3 meningioma line with catalogued mtDNA and a drug list

The widely used malignant meningioma line IOMM-Lee is confirmed NF2-wildtype with a pathogenic TERT promoter c.-124C>T variant. The authors add mtDNA variants, short doubling time (19–21 h), high colony-forming efficiency (up to 87%), relative radiosensitivity (D10 ~3.9 Gy) that ATM inhibition (AZD-1390) can worsen, and a 349-compound screen that yields 34 sub-micromolar clinical-use hits. This is a model-characterization resource, including mitochondrial genotypes, not a mitochondrial-mechanism paper.

Finding. IOMM-Lee, the workhorse Grade 3 meningioma line, is NF2-wildtype, TERT-promoter mutant, fast, clonogenic, relatively radiosensitive, and newly equipped with an mtDNA variant list and a 34-drug nanomolar hit sheet. ATM blockade makes radiation worse. It is related to other meningioma models and also distinct. Use the passport; do not pretend this is a mitochondrial-disease paper.

Why this paper matters

Bad models waste drug screens. IOMM-Lee was partly characterized. Filling exome, mtDNA, kinetics, radiation, and a clinical-compound deck makes the line safer to interpret. The TERT-124 / NF2wt combination locates it in a real genomic meningioma bin.

What to do with it

If you use IOMM-Lee, read the mtDNA and TERT calls before you claim an NF2 mechanism. Pull the hit list. Measure mitochondria if you care; they did not. The directional implication is that this malignant meningioma model is now genomically and pharmacologically better pinned, mtDNA included.
