Gelsolin keeps macrophage mitochondria intact during Legionella infection and buys survival without changing bacterial load

Gelsolin-knockout mice die faster from severe Legionella pneumophila pneumonia despite unchanged lung bacterial loads. Macrophages overproduce CXCL1/KC, lungs accumulate neutrophils and apoptotic cells, gelsolin-null neutrophils make fewer NETs and lose mitochondrial capacity, and infected macrophages lose mitochondrial network morphology and respiration. Recombinant gelsolin extends survival, reduces lung pathology, and calms the macrophage inflammatory signature. Host mitochondrial integrity, not CFU, is the survival variable.

Finding. Mice without gelsolin die faster from severe Legionella pneumophila pneumonia even though lung bacterial loads match wild type. Their macrophages overproduce CXCL1/KC, their lungs fill with neutrophils and apoptotic cells, their neutrophils make fewer NETs and lose mitochondrial capacity, and their infected macrophages lose mitochondrial network shape and respiration. Injecting recombinant gelsolin buys survival, cleans up lung pathology, and calms the macrophage inflammatory signature.

Why this paper matters

Legionnaires' disease is under-diagnosed and treated late. The usual therapeutic fantasy is a faster antibiotic. Whitham, Eltobgy, Shamseldin and the Amer group argue for a host-directed one: keep the phagocyte mitochondrion intact while the bacterium is still there.

The decisive design choice is CFU-independence. If gelsolin were an antimicrobial restriction factor, knockout lungs would carry more bacteria. They do not. The mice still die sooner. That moves the phenotype from pathogen control to immunopathology with a mitochondrial handle.

The organelle-to-inflammation chain

Gelsolin is required for mitochondrial network morphology and respiration in L. pneumophila-infected macrophages. That is the cell-intrinsic mitochondrial claim. Neutrophils lacking gelsolin produce fewer NETs and show diminished mitochondrial capacity in response to the same bacterium — a second phagocyte, same organelle theme. Upstream of tissue damage, gelsolin-null macrophages secrete more CXCL1/KC, and knockout lungs accumulate neutrophils and apoptotic cells.

The rescue is not genetic. Recombinant gelsolin protein given to knockout mice extends survival, reduces lung pathology, and, in vitro, reduces the macrophage inflammatory signature. That is stronger than a correlation. It does not yet say which gelsolin pool — cytoplasmic actin severing versus circulating plasma gelsolin — does the work in a wild-type host.

How to read the score

High mitochondrial and translational signal for an infection paper: morphology plus respiration in macrophages, mitochondrial capacity in neutrophils, in vivo survival, and a protein rescue. Confidence is high for the knockout phenotype and medium for mechanism. The abstract does not specify how gelsolin talks to fission/fusion machinery, which respiratory complexes fail, or whether wild-type mice plus antibiotics benefit. Do not file this as a Legionella virulence-factor screen.

What to do with it

If you track host-directed pneumonia therapy, gelsolin is now a mitochondrial-integrity drug, not only an actin-scavenger footnote. If you track mitochondria in innate immunity, pair this with other actin-regulatory proteins that gate mitochondrial dynamics during intracellular bacterial infection. The experiment that would move the score is recombinant gelsolin on top of standard antibiotics in wild-type animals, with a macrophage-autonomous mitochondrial rescue as the pharmacodynamic readout. Matched CFU is the control you should keep demanding.
